Simplex 2x12 Port Length

If one of my ports is 3/8” short, will that affect the sound? If so, how?

The cab will be tuned a bit high, but if it's only one of the six very slightly.

Would it be enough to make an audible difference? I’m wondering if I should get more material to make the proper port length.

You wouldn't hear it. You'd barely be able to measure it with laboratory grade equipment.
